Trên hình 3.10 cho thấy CHI từ giống đâ ̣u xanh và 10 mẫu phân tích được chi làm 2 nhánh chính. Nhánh chính thứ nhất bao gồm CHI của ngô, nho và CHI2 thuộc họ đậu hoang dã trong tự nhiên. Nhánh thứ 2 bao gồm các mẫu còn lại, CHI1, CHI2 từ đậu hoãng dã; cỏ linh lăng, sắn dây, cam thảo, đậu đỏ và đậu xanh. Trong đó, CHI từ đậu tương và cam thảo có hệ số
tương đồng di truyền cao nhất (96%). CHI từ đậu xanh có hệ số tương đồng cao nhất với CHI từ đậu đỏ (95%).
KẾT LUẬN VÀ ĐỀ NGHỊ
1. Kết luận
1.1. Gen mã hóa chalcone isomerase (CHI) đã được phân lâ ̣p thành công từ DNA genome và từ mARN tổng số của giố ng đâ ̣u xanh ĐXĐP và ĐXHL10. Kích thước gen CHI từ genome là 1110bp với 3 intron và 4 exon. Trình tự ORF của CHI có kích thước là 669 nucleotide, mã hóa cho 222 amino acid.
1.2. Hệ số tương đồng về trình tự nucleotide của gen CHI ở hai giống đậu xanh ĐXĐP và ĐXHL10 là 100%. Trình tự nucleotide gen CHI từ 2 mẫu nghiên cứu so với trình tự gen CHI ở đậu xanh mang mã số NM_001317294.1, KP164975 trên Ngân hàng Gen là 99,85% và không thấy có sự sai khác trình tự amino acid.
1.3. Trình tự amino acid CHI có tính bảo thủ cao ở thực vật, CHI của đậu xanh có sự tương đồng cao nhất so với đậu đỏ (95%).
2. Đề nghị
Tiếp tục nghiên cứu biểu hiện gen CHI nhằm tạo cây đậu xanh chuyển gen có hàm lượng Isoflavone cao.
