QUY TRÌNH ĐIỀU CHẾ HỆ VI TỰ NHŨ (SMEDDS) CHỨA TELMISARTAN Phạm Nguyễn Tường Vân Người hướng dẫn: TS Huỳnh Trúc Thanh Ngọc TÓM TẮT Mở đầu: Telmisartan (TMS) thuốc gây ức chế thụ thể angiotensin II sử dụng rộng rãi để điều trị tăng huyết áp phịng ngừa đột quỵ, có độ tan nước kém, dẫn đến độ hòa tan giới hạn sinh khả dụng thấp Đối tượng phương pháp nghiên cứu: Đánh giá độ tan TMS nhằm chọn tá dược, xây dựng giản đồ ba pha hóa muối phần để cải thiện độ tan TMS Các hệ SMEDDS đánh giá độ bền nhiệt động điều kiện môi trường khác Hệ sau tải TMS đánh giá độ bền nhiệt động, kích thước giọt, điện zeta độ hòa tan Kết quả: Đề tài xây dựng cơng thức quy trình điều chế hệ vi tự nhũ tải TMS 5% TMS hóa muối phần cách sử dụng dung dịch Kali hydroxid với nồng độ thích hợp Cơng thức cho 5g SMEDDS gồm 0,25g TMS, 1,38g Labrafac PG, 2,3g Kolliphor RH40, 0,92g Transcutol HP, 0,1g Kali hydroxid khan 0,05g Nước SMEDDS thu suốt, đồng nhất, có kích thước tiểu phân trung bình mơi trường nước 76,82nm, điện zeta -46,4mV bền vững mơi trường pH khác Tốc độ hịa tan TMS từ hệ SMEDDS- TMS thỏa yêu cầu USP 41 tương đương thuốc đối chiếu SMEDDS-TMS hóa rắn thành công với Florite R Viên nén thử nghiệm đạt u cầu tính chất lý có khả GPHC cao so với viên đối chiếu Kết luận: Đề tài bước đầu điều chế thành cơng hệ SMEDDS tải 5% TMS thỏa tính chất lý hóa Hóa rắn thành cơng với Florite R bào chế viên nén có độ hịa tan cao viên đối chiếu Quy trình điều chế có tính lặp lại, giải pháp tiềm giúp cải thiện độ tan, nâng cao sinh khả dụng Từ khóa: Hệ vi tự nhũ, SMEDDS, telmisartan iv Final thesis for the Master degree of Pharmacy – Academic year: 2018 – 2020 FORMULATION OF SELF MICROEMULSIFYING DRUG DELIVERY SYSTEM (SMEDDS) CONTAINING TELMISARTAN Pham Nguyen Tuong Van Supervisor: Huynh Truc Thanh Ngoc, Ph.D ABSTRACT Background: Telmisartan (TMS), an angiotensin II receptor inhibitor and widely used for the treatment of hypertension and stroke prevention, has a very low aqueous solubility which relates to limited dissolution and low bioavailability Methods: Telmisartan solubility was evaluated in order to select appropriate excipients, partial salt to improve the solubility of TMS and to construct phase diagram SMEDDS was evaluated for thermodynamic properties SMEDDStelmisartan was tested for thermodynamic properties, droplet size, zeta potential and dissolution properties Results: The present work has developed the formulation and the manufacturing process of SMEDDS loading 5% TMS in which TMS is incompletely neutralized by potassium hydroxid solution with a appropriate concentration The formulation containing 5g SMEDDS consists of 0.25g TMS, 1.38g Labrafac PG, 2.3g Kolliphor RH40, 0.92g Transcutol HP, 0.1g Dehydrated potassium hydroxid and 0.05g water The obtained SMEEDS, which is clear and homogeneous, has average particle size about 76.82nm in water, zeta potential value -46.4mV and shows good stability in various pH mediums The dissolution rate of TMS from SMEDDS-TMS formulation satisfied USP 41 and were comparable to the reference drug SMEDDS-TMS was successfully solidified with Florite R The tested tablet met the mechanical properties requirements and had a higher ability to release the active substance than the reference tablet Conclusions: The present work has achieved the formulation of SMEDDS loading 5% telmisartan, satisfied all physico-chemical
