Qui trình thực hiện
Bước 1: Truy cập vào trang web NCBI
Bước 2: Tiếp đó tại thanh tìm kiếm ta nhập các accession mà ta cần tìm Bên trái thanh tìm kiếm ta chuyển từ chế độ tìm “All Databases” sang chế độ “Nucleotide” Sau đó chọn Enter

Bước 3: Sau đó tại cửa sổ trang mới ta chọn “FASTA” để lấy các tình tự accession.

BLAST VÀ XÁC ĐỊNH TÊN LOÀI TỪ MẪU 4.3
Qui trình thực hiện
Bước 1: Trên thanh tìm kiếm của google, nhập từ khoá “Chromas” để tìm và tải ứng dụng.

Bước 2: Sau khi đã tải, ta mở ứng dụng Chromas lên

Bước 3: Tại giao diện màn hình chính, chọn Open rồi mở fide mẫu 4.3 lên.

Bước 4: Sau đó ta chọn “Export” Ở mục “ Save as type” ta chọn “Fomatted text” , chọn vị trí lưu fide rồi chọn “Save” Sau khi lưu, ta truy cập tiến hành xoá những đoạn nhiễu, ở phần đầu ta xoá 60 nucleotide, ở đầu cuối ta xoá 87 nucleotide

Bước 5: Truy cập từ khoá “blast ncbi” trên goolgle để tìm kiếm hoặc truy cập vào https://blast.ncbi.nlm.nih.gov/Blast.cgi để vào công cụ blast.

Bước 6: Tại giao diện màn hình chính ta chọn “Nucleotide BLAST”

Bước 7: Tại mục “chọn tệp” ta chọn fide của mẫu 4.3 Đặt tên mẫu tại mục “Job Title” Tại mục “optimize for” ta chọn “Optimize forSomewhat similar sequences (blastn)” Sau khi thiết lập hoàn tất ta chọn “Blast”.

Tỷ lệ Query Cover gần như 100% và chỉ số Per Ident cao cho thấy rằng trình tự DNA mẫu của chúng ta có sự tương đồng rất cao với các trình tự tham chiếu trong cơ sở dữ liệu Giá trị E cho tất cả các trình tự tham chiếu đều là 0.0 Điều này có nghĩa là khả năng ngẫu nhiên tìm thấy một kết quả tốt hơn kết quả truy vấn là rất thấp.Vì tất cả các trình tự tham chiếu đều thuộc chi Bacillus và thiếu thông tin để xác định loài cụ thể, ta có thể kết luận rằng mẫu DNA này là Bacillus sp.

Qui trình thực hiện
Bước 1: Trên thanh tìm kiếm của google, nhập từ khoá “mega x software” để tìm và tải ứng dụng.

Bước 2: Sau khi tải, truy cập vào ứng dụng MEGA

Bước 3: Tại giao diện màn hình chính của MEGA, ta chọn “Align” , sau đó chọn “ Edit/ Build Alignment”.

Bước 4: Một cửa sổ nhỏ hiện ra, ta chọn “Create a new alignment”, sau đó chọn “OK”.
Tiếp tục chọn “DNA” trong bảng mới xuất hiện sau đó

Bước 5: Tại giao diện cửa sổ mới ta chọn “Edit”, sau đó nhấn chọn “Insert Sequence From File” Tại đây, chọn fide Text document có chứa các sequence của trình tự mẫu 4.3 và các sequence của các accession number đã tìm ban đầu cần so sánh.

Bước 6: Xoá Sequence trống cuối Sau đó chọn “Alignment”, sau đó chọn “Alignment by ClustaIW”.

Bước 7: Chọn “Data” , chọn “Export Alignment” để xuất fide, tiếp tục chọn định dạng của file là “MEGA format” Sau đó chọn “Save” để lưu thư mục

Step 8: In the MEGA software interface, select the icon labeled "Phylogeny", then select "Construct/Test Neighbor - Joining Tree " Choose the previously saved MEGA fide, then select "OK".
