truyền nhiễm gà (Chicken Infectious Anemia Virus - CIAV) Hà Nội vùng phụ cận” Ngành: Thú y Mã số: 60.64.01.01 Tên sở đào tạo: Học viện Nông nghiệp Việt Nam Mục đích nghiên cứu: - Xác định có hay khơng có lưu hành CIAV đàn gà thuộc địa bàn nghiên cứu - Nghiên cứu số đặc điểm dịch tễ học phân tử CIAV Nội dung nghiên cứu Trong khuôn khổ nghiên cứu đề tài, thực nội dung sau: - Xác định lưu hành CIAV đàn gà nuôi Hà Nội vùng phụ cận - Giải trình tự gen mã hóa protein VP1 CIAV - Phân tích số đặc điểm dịch tễ học phân tử CIAV Phương pháp: Phương pháp lấy mẫu; Phương pháp tách chiết ADN; Phương pháp tối ưu hóa phản ứng PCR chẩn đoán CIAV; Phương pháp nghiên cứu đặc điểm dịch tễ học phân tử Kết kết luận: (1) Trong số 14 trại kiểm tra có đến 13 trại (tỷ lệ 92,86%) phát có lưu hành CIAV; 01 trại (tỷ lệ 7,14%) âm tính với kết PCR phát CIAV Trong tổng số 124 mẫu bệnh phẩm gà có triệu chứng lâm sàng kiểm tra tỷ lệ phát CIAV cao (63 mẫu dương tính, tỷ lệ 50,81%) (2) Đã giải trình tự gen thành công phân đoạn gen VP1 CIAV 26 chủng CIAV lựa chọn để giải trình tự có mức tương đồng cao trình tự nucleotide, từ 96,8% đến 100% (3) Qua phân tích đặc điểm dịch tễ học phân tử dựa vào trình tự gen mã hóa protein VP1, kết luận: - Các chủng CIAV lưu hành có đa dạng tương đối mặt di truyền - Các chủng nằm nhánh di truyền khác với chủng virus vacxin viii THESIS ABSTRACT Name of Master student: Nguyen Quoc contents: - Determining the prevalence of CIAVof the chicken in Hanoi and surrounding areas - Sequencingprotein VP1 geneof CIAV - Researching molecular epidemiological characteristics of CIAV Methodology - Sampling method; - DNA extraction method; - PCR for identifying CIAV; - Methods for researching molecular epidemiological characteristics of CIAV Results: (1) By using PCR,13 farms (92.86%)of tested 14 farms were CIAV positive; there was only one farm (7.14%) was CIAVnegative; In total124 samples of chickenwhich had clinical symptoms,there were 63 CIAV positive samples (50.81%); (2) 26 sequencing of VP1gene of CIAVin this study sharedfrom 96.8% to 100% nucleotide identity (3) By phylogenetic analysis based on nucleotide sequences of VP1 gene of CIAV showed that: - CIAV strains in this study have geneticvariation; - These strains were located in different branches of phylogenetic tree with vaccine virus strains ix
