dựa cấu trúc sợi nano Platin để định lƣợng cholesterol dung dịch” đƣợc thực từ tháng 01/2012 đến tháng 07/2012 Phòng thí nghiệm Cơng nghệ Nano – Đại học Quốc gia Thành phố Hồ Chí Minh với vật liệu nghiên cứu cảm biến sinh học sợi nano Platin enzyme cholesterol oxydase Khóa luận gồm hai nội dung nghiên cứu Nội dung cố định enzyme lên sợi nano Platin Trong nội dung này, trình làm bề mặt cảm biến quy trình biến đổi bề mặt cảm biến với trình tự lớp đơn tự ghép cysteamine, chất hữu trung gian glutaraldehyde, enzyme cholesterol oxydase đƣợc khảo sát Nội dung nghiên cứu ứng dụng cảm biến đƣợc cố định enzyme để kiểm tra định lƣợng nồng độ cholesterol từ 0,8 – mM dung dịch PBS phƣơng pháp quét dòng tuần hồn (cyclic voltammetry - CV) Kết trình xây dựng quy trình biến đổi bề mặt chip cố định enzyme cho thấy nồng độ dung dịch cysteamine tối ƣu 0,5 M; thời gian phản ứng 40C nồng độ dung dịch glutaraldehyde tối ƣu 1%; thời gian phản ứng 40C Cố định thành công enzyme cholesterol oxydase lên cảm biến sau đƣợc biến đổi bề mặt enzyme giữ đƣợc hoạt tính sau cố định Kết trình kiểm tra định lƣợng cholesterol nồng độ khác dung dịch PBS cảm biến sau cố định enzyme cho thấy ngƣỡng phát cholesterol 0,8 mM, xác định đƣợc đỉnh oxy hóa – khử +0,625 V Quá trình kiểm tra định lƣợng cholesterol cho thấy có mối tƣơng quan tuyến tính chặt chẽ cƣờng độ dòng thu đƣợc nồng độ cholesterol khác theo phƣơng trình Ipeak = 13,97*Ccholesterol + 221 với hệ số tƣơng quan R2 = 0,974 Với kết đạt đƣợc, đề tài “Nghiên cứu cố định enzyme cholesterol oxydase cảm biến nano sinh học dựa cấu trúc sợi nano Platin để định lƣợng cholesterol dung dịch” bƣớc khởi đầu cho trình chế tạo thiết bị cảm biến định lƣợng cholesterol máu ngƣời ii SUMMARY The thesis “Study on the immobilization of cholesterol oxydase on bionanosensor structure based on Platinum nanowires to quantify cholesterol in solution” was carried out at laboratory for Nanotechnology National University Ho Chi Minh city from January 2012 to Junly 2012 with the main materials are Platinum nanowires and cholesterol oxydase The thesis has two main subjects The first, immobilization of enzyme on Platinum nanowires In this part, the process of cleaning sensor suface and process modified electrodes suface with cysteamine self-assembled monolayer, the intermediates organic glutaraldehyde and enzyme cholesterol oxydase will be investigated In the next part, use sensors applications have been fixed with enzyme cholesterol oxydase to check, quantify the cholesterol has concentration from 0,8 to mM in PBS solution by using Cyclic Voltammetry mode in Autolab machine The resulfs were summarized as follows: Complete the process modified electrodes suface through cysteamine self- assembled monolayer and organic glutaraldehyde with the best solution concentration cysteamine is 0,5 M, it react in two hours at 40C The best solution concentration glutaraldehyde is one percent, it react in two hours at 40C The cholesterol oxydase enzyme was immobilized successfully on the modified surface of Platinum nanowires electrode Checking and building successfully standard equation The regression equation for cholesterol concentrations and its relevant oxydation current reponse was Ipeak = 13,97*Ccholesterol + 221 with an approximate correlation of R2 = 0,974 and sensor sensitivity of 0,8 mM The experiment results also express the
