NGHIỆP LỜI MỞ ĐẦU Đặt vấn đề Những năm gần đây, nghề nuôi trồng thủy sản không ngừng phát triển ngày có vị trí quan trọng kinh tế Việt Nam Đồng Bằng Sông Cửu Long (ĐBSCL) vùng đất giàu tiềm thiên nhiên ưu đãi có lợi lớn để phát triển nuôi trồng thủy sản Nuôi trồng thủy sản ĐBSCL có bước phát triển vượt bậc mang lại hiệu kinh tế cao, đặc biệt cá tra đối tượng nuôi chiến lược, thu lại nguồn ngoại tệ lớn, xem nguồn xuất thủy sản quan trọng nước ta Tuy nhiên với gia tăng mức phong trào nuôi cá tra ĐBSCL, tạo điều kiện cho dịch bệnh thường xuyên bộc phát gây nhiều thiệt hại cho người nuôi Trong đó, vi khuẩn coi tác nhân gây bệnh nghiêm trọng gây tỉ lệ hao hụt cao cá tra Đặc biệt vi khuẩn Aeromonas hydrophila gây bệnh xuất huyết tác nhân gây bệnh làm tổn thất lớn cho người nuôi năm Vi khuẩn A hydrophila xem đa dạng di truyền nghiên cứu nước di truyền vi khuẩn hạn chế Sự phân loại chi Aeromonas thường phức tạp, khó khăn việc nhận biết kiểu hình Aeromonas sử dụng phương pháp khác nhau, điều kiện test sinh hóa, biến đổi kiểu hình loài Ngày nay, với phát triển hiểu biết sinh học phân tử, nhiều thị sinh học phân tử nghiên cứu ứng dụng công cụ hỗ trợ đắc lực cho công tác nghiên cứu khả gây bệnh chủng A hydrophila Để góp phần xác định đa dạng di truyền chủng gây bệnh A hydrophila, tìm liên hệ kiểu gen độc lực vi khuẩn nhằm có đươc biện pháp hiệu hạn chế thiệt hại bệnh gây ra, đề tài “Sự khác biệt di truyền chủng Aeromonas hydrophila phân lập cá tra (Pangasianodon hypophthalmus) Đồng Bằng sông Cửu Long” thực Mục tiêu nghiên cứu: Đánh giá khác biệt di truyền chủng A hydrophila phân lập cá tra ĐBSCL phương pháp sinh học phân tử ĐỒ ÁN TỐT NGHIỆP Nội dung nghiên cứu: Khảo sát diện gen gây độc A hydrophila phương pháp PCR xác định đa dạng protein tổng số vi khuẩn 18 chủng A hydrophila phân lập cá tra Giải trình tự đoạn gen 16S rRNA rpoD 18 chủng vi khuẩn để khẳng định lại định danh lồi kiểm tra sinh hóa, đồng thời thiết lập phát sinh lồi tìm mối tương đồng di
