Các kí hiệu phổ s: vân đơn d: vân đôi t: vân ba dd: vân đôi đôi brs: vân đơn tù m: vân bội - Các kí hiệu khác KB: Tế bào ung thư biểu mô MCF7: Tế bào ung thư vú SK-LU-1: Tế bào ung thư phổi Hep-G2: Tế bào ung thư gan Hela: Tế bào ung thư cổ tử cung SK-Mel-2: Tế bào ung thư da AGS: Tế bào ung thư dày IC50: Nồng độ ức chế 50% LD50: Liều chết 50% động vật thí nghiệm ED50: Liều hiệu 50% đối tượng thử A: Acetone E: Ethyl acetate H: n-hexane MeOH: Methanol CTPT: Công thức phân tử MỞ ĐẦU Ơ nhiễm mơi trường dẫn đến việc phát sinh loại bệnh dịch bệnh ung thư, AIDS, COVID-19… chưa tìm thuốc đặc trị Trong công b ố T ổ chức Y t ế gi i, năm 2020 tồn cầ u có 19,3 tri ệu ngườ i mắ c ung thư, ,96 tri ệu ngườ i ch ết ung thư H ằ ng năm Việ t Nam có kho ả ng 70000 ngườ i ch ế t b ệnh ung thư có kho ả ng 200000 ca mắ c mớ i Do đó, việ c tạ o lo i thu ốc mớ i, có hiệ u qu ả chữa tr ị nhiề u lo ại b ệ nh t ật điề u c ầ n thi ết Để đáp ứng nhu c ầ u đó, vi ệ c nghiên c ứu hóa th ực vậ t đượ c ý, thu hút s ự quan tâm rộng rãi nhà khoa học Điển việc tìm taxol từ Thông đỏ, vinblastine vincristine từ Dừa cạn…ứng dụng làm thuốc chữa ung thư Việt Nam quốc gia có tính đa dạng sinh học cao với gần 12000 loài thực vật bậc cao thuộc 2256 chi, 305 họ (4% tổng số loài, 15% tổng số, 75% tổng số họ thực vật lớn) Hệ thực vật rừng khơng mệnh danh xanh điều hịa khí hậu mà cịn mang đến tiềm to lớn thuốc đóng vai trị nịng cốt việc bảo vệ sức khỏe hạnh phúc cộng đồng giới Với số lượng lớn vậy, nhiều thuốc xa lạ với người dân người dân chưa có kiến thức đầy đủ tác dụng cách sử dụng Trong tìm hiểu loại thảo dược lưu truyền từ đời sang đời khác Việt Nam, nhân dân ta sử dụng loài thực vật họ Trôm (Sterculiaceae) để điều trị hiệu nhiều bệnh khác Trong dân gian, An xoa (Helicteres hirsuta) thuộc chi Thâu kén họ Trôm (Sterculiaceae) sử dụng để điều trị bệnh kiết lỵ, bệnh sởi, Một ứng dụng đáng ý khác loại chữa bệnh liên quan đến gan tốt Tuy nhiên, nghiên cứu hóa thực vật chúng hạn chế Cây Màng kiêng (Pterospermum truncatolobatum) số lồi thuộc chi Pterospermum họ Trôm (Sterculiaceae) phát Việt Nam Cho đến nay, Màng kiêng nhắc đến phân loại thực vật Trên giới, số hoạt chất phân lập từ loài thuộc chi Helicteres chi Pterospermum Tại Việt Nam, cơng trình nghiên cứu hóa dược hai 114 Hợp chất M3 quan sát tạo liên kết hydro với Arg59 His63, bên cạnh đó, liên kết phối tử protein củng cố bền vững thêm thông qua liên kết không cực với Arg69 Ala74 Hợp chất L5 hình thành liên kết hydro với amino acid Asn65, Val67, Arg69 Các liên kết yếu tạo thành với Leu62 His63 góp phần tăng cường lực liên kết hợp chất với đích protein nghiên cứu Kết sở khởi đầu, định hướng cho nghiên cứu sâu chất ức chế protein wMUS81 mơ hình thực nghiệm in vitro in vivo 115 KẾT LUẬN Sau thời gian nghiên cứu thực đề tài, đạt kết nghiên cứu sau đây: Về thành phần hóa học: - Cây An xoa: Đã phân lập xác định cấu trúc hóa học 14 hợp chất, có hợp chất từ rễ (L1÷L9), hợp chất từ thân (L10-L14) An xoa Đó methyl betulinate (L1), stigmasterol (L2), 3β-benzoylbetulinic acid (L3), methyl 3,4dihydroxybenzoate (L4), 3β-O-trans-caffeoylbetulinic acid (L5), p-hydroxybenzoic acid (L6), β-amyrin (L7), 3,5-dimethoxy gallic acid (L8) betulinic acid (L9), icosanoic acid (L10), 3β-hydroprotosta-17(20),24-dien-21-oic acid (L11), 5,7dihydroxy-4’-methoxy-flavone-8-yl O-β-D-glucopyranoside (L12), methyl caffeate (L13), 5,8-dihydroxy-4’,7-dimethoxy-flavone (L14) Trong đó, hợp chất 3βbenzoylbetulinic acid (L3) chất - Cây Màng kiêng: Lần phân lập xác định cấu trúc hóa học hợp chất Đó β-sitosterol (M1), icosanoic acid (M2), bentulonic acid (M3), simiarenol (M4), stigmasterol (M5), stigmasterol glucoside (M6), β-taraxerol (M7), 2-methoxy benzoic acid (M8) Trong đó, cấu trúc M4 M7 xác định X-ray đơn tinh thể Về hoạt tính sinh học Đã tiến hành thử hoạt tính chống oxi hóa, hoạt tính kháng vi sinh vật Một số hợp chất phân lập tiến hành thử khả gây độc tế bào dòng ung thư Các kết thu cho thấy: - Cao phân đoạn EtOAc mẫu thân - rễ có hoạt tính chống oxi hóa mạnh với IC50 tương ứng 0,112 0,117 mg/ml - Kết thử nghiệm khả ức chế dòng tế bào ung thư biểu mô KB cho thấy: cao chiết từ An Xoa (TLE, RH, RE, RB) cao chiết (PH, PD, PE PB) từ Màng kiêng có hoạt tính (IC50 dao động từ 7,4 đến 182,51 μg/ml) Trong đó, cao chiết EtOAc rễ An xoa Màng kiêng có khả gây độc tế bào ung thư dòng KB mạnh với IC50 3,23 µg/ml 7,6 µg/ml -
