Các bước thực hiện
Bước 1: Truy cập vào trang web NCBI (https://www.ncbi.nlm.nih.gov/)
Mở list All Databases chọn Nucleotide, Nhập Accession Number + 16S rRNA (Ví dụ: “KP699121.1 16S rRNA”) Nhấp Search.

2.2.2 Blast và xác định tên loài của mẫu 1
Bước 1: Lưu file mẫu về thiết bị.
Bước 2: Tải Chromas (http://technelysium.com.au/wp/chromas/) và khởi động phần mềm.

2.2.3 Xây dựng cây di truyền các accession number
Bước 1: Mở chương trình Mega.
Bước 2: Chọn “Align” sau đó chọn “Edit/Build Alignment”.

Bước 3: Cửa sổ “Alignment Editer” hiện ra, chọn “Create a new alignmet”, sau đó nhấn “OK”.

Bước 4: Một cửa sổ hiện câu hỏi “Are you building a DNA or protein sequence alignment?”, chọn “DNA”.

Bước 5: Giao diện mới hiện ra Sao chép các trình tự 16s rRNA của mẫu 1 vào chương trình.

Bước 6: Ctrl+A để chọn tất cả các trình tự Chọn “Alignment”, sau đó chọn

Bước 7: Cửa sổ mới hiện ra chọn “Neighbor Joining” trong “Cluster Method” và nhấn “OK”

Bước 8: Kết quả hiện ra chọn “Data” ► “Export Alignment” ► “Mega Format” Lưu file lại với đuôi “.MES
Bước 9: Sau khi lưu thì đóng trang và quay lại trang đầu tiên chọn “File” ►
“Open A File/Sesion”, dẫn đến file vừa lưu ở Bước 8
